Now that sounds like a credentials issue. Who is the database owner? Did you use an object_qualifier for the DNN setup in the Web.config. What login are you using to access SQL DB?
Provided you're not using "Integrated Security (aka- Network Service)", try using that account to access the DB through the Management Studio. Also, under Security\Logins, see if the "sa" account is enabled (if it's not it should have a little red arrow next to the icon.). After enabling it, try logging in as 'sa'.

Do you need to set objectQualifier to something under "SqlDataProvider" in order to access the DNN database using Sql Server Management Studio Express?

no, oq is only used to identify objects of one dnn installation within Db.

with SQL Express you can have file based connection to the database (default DotNetNuke configuration) and need to be careful, accessing the database using Management suite, see http://www.dotnetnuke.com/Community/Blogs/tabid/825/EntryID/1202/Default.aspx for details.
