Hi - I did this recently. To play it safe I first upgraded to 3.3.1, then I switched the web site to ASP.NET 2.0, and then I upgraded 3.3.1 to 4.3.1.

Basically, for both upgrades (3.2.2 to 3.3.1, and then 3.3.1 to 4.3.1) I just downloaded the upgrade package, unzipped on my harddrive, applied changes to web.config, uploaded over the existing installation, and ran the install utility.

I've completed that same sequence for a few installations, you should be able to do it no problem.
