I tried to enable composite files but it seems no effect for my site. Is it because my site is using multi-language (en-us and ja-jp)? If so, can you kindly advise me how to resolve such issue for my multiple-language site?

I am not aware of limitations with combined files in multilingual sites.
Which DNN version are you using and how did you detect, that there are no changes.

Product: Evoq Content Basic 

Version: 08.01.00 (795)

I have two sites to test. One is multi-language portal while another is single language portal. 

After enabling composite files for each site and viewing source, the single-language portal showed "/DependencyHandler.axd" which I assumed has composited files successfully. In multi-language portal, I still don't see any changes in its source. See the screenshot.

Does your web.config file have debugging enabled? I think that turns off composite files.

I did make sure that web.config's debug mode is off. See below:-
