We have an existing setup of DNN 2 which is using SQL server 2000.
Would there be any issues if we move the DNN 2 database to SQL server 2005 on another machine?

SQL 2005 is mainly backward compatible with SQL 2000 - it has been reported, that a few SQL statements are not interpreted exactly the same, so you might need to make minor tweaks to a few stored procedures.

I'm running approximately 10 sites ranging from DNN 1.09 to DNN 3.3 with sql server 2005 with no issues (zero / zilch / nada).

If however - you are migrating to windows 2003 / ii6 - I had major stability issues with early versions of DNN that were only resolved when I set IIS to run in ISS 5 isolation mode. I initially though sql server 2005 was the culprit - but eventually pegged it to IIS after one very stressful week...
