Rob are you getting any sort of Javascript error? I just want to rule that out first. Also, have you tried from a different computer (to rule out anything on your computer)?

I've encountered the same issue with my DNN 4.3.5 installation.  I have several pages built using Core and Non-Core modules.  I attempt to edit the Settings of a particular core module and the UPDATE simply does not respond.  It is as if the link is disabled or something.  It is so frustrating.

I have the same issue with DNN 4.3.4. I discovered that if you click first on the green arrow placed at the right of Categories list it works. I experienced also that it happens if you're using Mozilla browser. With MSIExplorer everything works fine.
