When I try to autogenerate the zip file with the module definitions, create private assembly option I get the following error:

Access to the path 'C:\datos\projects\DotNetNuke\Website\DesktopModules\GestorRestricciones\GestorRestricciones.dnn' is denied.

The weird thing is that I get the error for a couple of my custom modules, another couple generate the zip file just fine.

What can be wrong?

Check you dnn file is not under source control or somehow Wrie Protected.  Part of the packaging routine updates the dnn file.
