To manually make a localized resource file, try this: 
Place a duplicate of the resource file you want to localize in the same folder but with a special name format that signifies the resource file is for use only by a particular portal.  The format inserts "Portal-<portalid>" between the file name and file extension.

Make a copy of  <resource-file-name>.resx   and name it   <resouce-file-name>.Portal-<portalid>.resx

Example for manually creating a localized GlobalResources file for use only by Portal 2: 

Copy  GlobalResources.resx  and name it  GlobalResources.Portal-2.resx

This method works for both module resource files as well as DNN core resource files.  Remember:  Make a COPY of the original resource file (leave that original alone!) and place the renamed copy in the same folder as the original.

Using the DNN admin interface is probably your safest bet but we do this manual method without issue on a pretty routine basis across both DNN 4.0.x and DNN 4.3.x sites.
