Which is the correct way of exporting a portal that is running on one DNN server and importing it on to another DNN server?

An installation consists of 2 things - the files on server disk and your database.  You will need to copy the DNN files to the new server and then restore the database backup on the new SQL server.  You will then need to edit the connection string for the new database location.  I'm leaving out things like proper IIS permissions etc...This is not the way to do it if the destination is a live site.

If the destination is a live site I'm not sure the best way.  You could try exporting the existing portal as a template via Host Settings - Portals (choose to include content) and then create a new portal with that template on the new site (upload template first).  I've never done this so I do not know how well it would work.  You will probably need to take into account any 3rd party modules in use at the source at the destination too. 

For the time and effort it would save you - maybe consider purchasing one of the commercial site back up solutions available at Snowcovered.  I think some advertise that they can restore too.
