Question 1
Can you give some ideas of hardware sizing / hosting requirement we would need for
 - 25K, 50K, 100K, 250K, 500K Members on the database
 
The usage pattern will be
 - UK/mainland Europe (day and early evenings)
 - access to simple web pages (HTMLText module)
 - view/update profile (core DNN module)
 - contribute to forum (Active Forums module)
 - download small documents <100K (core DNN Repository module)
 - assume 10% concurrency

....  plus recommended Hostign provider
Question 2.:
In your experience what is the maximum no of Members (held on the database and then concurrent) that DNN can cope with.

this is very difficult to guess, but DNN scales very well. You can start with a single server for Web Site and database, for performance and security reasons, I usually recommend to use separate machines for both. DotNetNuke.com has been using this configuration since the got more than ~300.000 registered users, and switched to using a web farm since then.
