Running Windows 7

SqlSvr Express 2008 installed

Passed all file Permissions test  but received following.  during Configure Dabase Connection

Select Database:  SQL Server 2005/2008 (Express) File

Server:    .\SQLEXPRESS

Filename:    Database.mdf

Integrated Security:   X

Run as db Owner:    X

Connection Error(s):
Index #: 0
Source: .Net SqlClient Data Provider
Class: 14
Number: 15382
Message: Failed to generate a user instance of SQL Server due to failure in retrieving the user's local application data path. Please make sure the user has a local user profile on the computer. The connection will be closed.

I would appreciate any direction that works to solving this connection issue

You need to run as a user account that has a local profile to use the automated connection string.  Not sure if IIS 7.5 is the same as IIS 7, but try going into the IIS Admin, select the application pool, click on Advanced and set LocalUserProfile to True.  I'm not a big fan of user instances in SQL and this is one of the reasons.
