Horacio, Charles Nurse (who is sitting next to me here at the ASP.Net insiders day in Redmond) just confirmed that in PortalLocalization table, there shouldn't be a row without a culture code. Please backup your database and delete it.

The problem is that I've already tried deleting this row. The problem remains the same.

To test what happens, I've deleted the row with blank CultureCode.
I've got the same error when I've edited the es-ES language:

Any other ideas on this? 

We have a lot invested in the site with hundreds of active users, so we need to move this forward.  Unfortuneately it is not a simple "roll back" issue.

Should we start debugging the core code?
