 This in my Connection String

===================================

<connectionStrings>

    <!-- Connection String for SQL Server 2005/2008 Express -->

    <add name="SiteSqlServer" connectionString="Data Source=.\SQLExpress;Integrated Security=True;User Instance=True;AttachDBFilename=|DataDirectory|Database.mdf;" providerName="System.Data.SqlClient" />

    <!-- Connection String for SQL Server 2005/2008

    <add name="SiteSqlServer" connectionString="Server=(local);Database=DotNetNuke;uid=;pwd=;" providerName="System.Data.SqlClient" />

connection string looks fine, but for some reason the database file cannot be opened, e.g. because you attached it to sql server.
If possible, use attached database instead, connectionstring should look like "Data Source=.\SQLExpress;Database=DotNetNuke;Integrated Security=True".
You'll need to create an empty database called "DotNetNuke" (or modify database in connection string), a SQL Server login for the AppPool Identity ("IIS AppPool\DNN, if DNN is name of your IIS application pool, which uses AppPool Identity) and grant dbowner permission to it.
