Mark, did you grant view permission for anonymous users in IIS virtual directory configuration as well?

Did you change security settings?

One question that you need to answer is whether or not the problem is with DotNetNuke or with your website vis a vis IIS.

To answer this question, drop a simple index.htm file into your root directory.  If you browse to the site, your browser should display that file.  If not, then you know that your problem is related to IIS.
