Welcome to DotNetNuke community.

Regarding your questions:

1. no, you need Microsoft SQL Server (any version 2000/2005/2008, including free Express )

2. you need Windows Hosting (IIS), there are detailed installation instructions available at mitchelsellers.com

Welcome to DotNetNuke!

Clarifying a point about "host"...
If by "host" you mean a local host (your own computer) then, as Sebastian mentioned, the video tutorial from Mitchel Sellers is a decent place to start.  Though it's for an older version of DNN (4.5.5) it is still fairly relevant.

If by "host" you mean an online hosting provider (GoDaddy, etc) then you may also have the option of "auto-installation" of DNN using your hosting provider's web management interface.  This is usually much easier than a manual instllation but such an installation may include other pitfalls.  There is info on using the DotNetNuke auto-installer on GoDaddy here and the auto-installer on Network Solutions here (including a link to an excellent PDF document from Chuck Self).
