I have spent all day trying to install DNN 4.0.3.  Every time the system report the following error.  Can someone experienced please help me sort this out?  Much obliged.

This issue has come up before. I suggest you post a message here and share your experiences. Hopefully, you can work together to find a solution. It's hard for us to help when we can't reproduce the problem.

I finally figured out what I was doing wrong.  I had the same error, so maybe you are making the same mistake.

Like you, I followed the instructions to the letter.  Never having used this product before this was a must!  Well what I figured out was using the examples you create a folder in the root of c:\.  Install  the software in it, set it as a virtual host location.  Then you go to visual studio and create a new web site.  Visual creates this site in your My Documents/visual studio 2005/websites directory by default.  Your web.config file does not exist in this directory, and your virtual location does not point here.

I simply renamed the release.config file in my documents directory and changed my virtual host location and all worked.

Maybe you have the same problem.
