I am getting the following error when i upgraded to 5.5.1. This error is when you go to Host-> Module Definitions:

this sounds like an upgrade did not run smoothly. Some of the controls have been moved, you may adjust paths inside Host :: Extensions.

Was your path initially configured to run at a URL like www.mydomain.com/dotnetnuke/ ? When you ran the upgrade process, did you make sure to UNBLOCK the ZIP file before you extracted it? 

I posted a new video yesterday on how to Upgrade DotNetNuke, it might be worth watching.
