OK, I loaded AnySQL Maestro onto the IIS server box and found that I had to add a SQL Authenticated user and was able to connect.

You should then be able to connect from dnn using the same connection settings. Westa

OK, the Query Builder's connection string is similar to this:

Provider=SQLOLEDB.1;Password=**********;Persist Security Info=True;UserID=DotNetNuker;InitialCatalog=DotNetNuke;Data Source=acsql

So, I want to set the two web.config file connection string entries to something like this:

<add name="SiteSqlServer" connectionString="Server=acsql;Database=DotNetNuke;uid=DotNetNuker;pwd=**********;" providerName="System.Data.SqlClient" />

ASP.NET version 2.0 or 4.0?

Anything else to set inside of IIS besides pointing to the DNN directory, enabling Anonymous access, and enabling Integrated Windows authentication?

When I try to launch the install page as http://localhost/dotnetnuke, what I get is a 404 error.
 
I don't see where the problem is.

In your DNS or HOSTS file how are your domains defined.
We normally setup a new DNS pointer for each hosting using localhost if not a good way to go.

dnnDEV.local  or some such

In IIS how are your bindings defined.
