I know this is probably a very simple problem but ... Have installed latest 4.3.4 on Win 2003 with SQL Server 2005. Have followed all installation instructions including granting rights to the DNN folder to NETWORK SERVICE. When I try to access the site to initiate the installation I get HTTP 404 error. So, DNN installed to D:\DotNetNuke\Website, Virtual IIS directory created pointing at this DNN directory (virtual directory called DotNetNuke), NETWORK SERVICE has all rights to D:\DotNetNuke and subfolders. I have created a simple html file which opens fine from http://localhost/DotNetNuke/test.html. Not sure what to do next. Anyone got any ideas!!

No idea but I am having the same issue.   So you are not alone!

I never encountered this problem when testing on Win XP.However, when setting up DNN on a 2003 server I got the exact same problem.

What did the trick for me is the following; Go to IIS mamagement console and enable .net 2.0 at the web extensions.

Hope this solves your problem too.

Just to clarify when you say initiate installation you are referring to

http://localhost/dotnetnuke/install/install.aspx?mode=install

correct?

I just ran into the same problem.  In my situation it was because asp.net 2.0 was not properly registered.  I re-registered it and all went fine.
