You've proven that you have not installed aspnet correctly.  Probably installed IIS after installing aspnet.

run "aspnet_regiis -i"

The earlier problems you mentioned about were probably permission problems. If your server is set up for testing purposes only and dont have connections to the internet, a surefire way to overcome them in my experience is to give access to your dnn folder i.e the physical and not the logical or virtual folder to the computer user 'everyone'. It should get it working then.

These settings when setting up the virtual directory should work:

I will often get an access denied error when I use Windows Explorer to setup my virtual directory.  The reason it occurs for me is because the directory is not setup to allow anonymous access.  In order to run DNN and develop DNN, every site requires two authentication types:  1) Anonymous access for use by anyone using the webbrowser and 2) Windows Authentication for use by VS.Net when loading the site project.  Both of these authentication settings need to be setup in IIS.

also, for a full overview of all access rights needed for asp.net to run, see this MSDN library article: http://msdn2.microsoft.com/en-us/library/kwzs111e.aspx
