I have tried this numerous times and get the same problem.  I create a new portal but when I go to the page I set as the alias, I get a Page Not Found error.

I am creating the portal on my local machine.  Is there some configuration that I need to do on IIS first?  My main portal displays fine with an alias of http://localhost/dnn but the new portal I'm tring to create with http://localhost/portal2 is not found even though I see the new subfolder in my portals directory.

Thanks in advance for any help that someone can send my way.

Have you created a Virtual Directory named "portal2" in your IIS? Let it point to the same directory (= the DNN root directory) as the first portal. DNN will use the alias to access the required portal.
