How do I specify a different email sender for each parent portal?

You don't.  Unfortunately.  I think there was a plan to change this in an upcoming version, but it doesn't appear to be in 5.0.

So the email address in the Host Settings --> SMTP Settings is the only email address that can be used for sending.

You cannot set each parent portal's sender email address to the administrator's email address of each parent portal somehow?

You can only specify one SMTP server in DotNetNuke.
Obviously your SMTP server must allow sending of emails from the email domain you are specifying in each portal.

There is an email account in Host Settings (as you specify), also check the Host superuser account email (Host/Superuser accounts).

Each portal has an admin account (Admin/User Settings) and you can specify the email address there for each portal.

Alex has it right.  You can specify a different admin email address for each portal but you can only specify one SMTP server for the entire DNN instance.
