There are a lot of issues which can arise during an installation and it is difficult from the information given so far to determine a cause or solution.  Probably the easiest method to install DotNetNuke locally is to use MakeDNNSite.  This application will walk you through the process of installing DotNetNuke with a minimum of hassle and should help isolate installation issues.

I managed to sort it out (sort of.) I took an installation off a colleague's work station, exported his IIS site as a package, did a SQL backup, and restored it all to my machine with a few tweaks for my environment. It worked.

(I then backed up everything I had set up in the same way, so I can restore easier if I blow it up again.)

It is always good to report a resolution so that others can resort to the same thing if they can, so there we go!
