I need to set up some skins/designs with custom modules to demonstrate what DNN can do fairly quickly ,so if any one can provide any links or tips I will be grateful.
1. Yes, you can do this via the Admin/Site Settings page, though if you have skins defined at the PAGE level they will override this setting.
2. Not sure exactly which question you are asking here

You can add a module to a page in a specific Pane (contentpane, leftpane, right pane, etc) then in the Module Settings you can choose Display On All pages, and that module will then be placed on every page of your website, in the same PANE on each page. If a particular page doesn't have that pane, it will end up in ContentPane because ContentPane exists in every skin. Not all panes are in every skin, it depends on the designer.
4. When creating a page you can choose a skin in the appearance section, which skin you choose decides how many columns are available. How many columns each skin has is something that was decided when the skin was created.
