I would like to create some custom profile fields for our internal users and then synchronize them with our HR database. Therefore in my synchronize-module I would need something like if (exists(profile["customField"])) { string customField = profile["customField"]; //etc. }

How are the correct functions for that?

If you want to determine if a custom profile property has been defined:

Dim ppd As ProfilePropertyDefinition = ProfileController.GetPropertyDefinitionByName(PortalId, "customField")
If ppd Is Nothing Then
    'Custom property has not been defined
Else
    'Custom property has been defined
End If

If you want to determine if custom profile property has been set in a particular user's profile:

Dim propValue As String=String.Empty
Dim ppd As ProfilePropertyDefinition = UserInfo.Profile.GetProperty("customField")
If ppd IsNot Nothing Then
    propValue = ppd.PropertyValue
    If propValue Is Nothing Then
        If Not String.IsNullOrEmpty(ppd.DefaultValue) Then
              propValue = ppd.DefaultValue
        End If
    End If
End If

In the second example above, this is one of several ways to obtain a profile property value.

Thanks, that's great! You define the propValue as a string. How would this be for e.g. a dropdown property value?
