This would make a fascinating study in versioning!  While researching the changes in various user related library code for a problem that I was having, I came across the solution to your problem.  I suspect that you first downloaded the 4.3.1 source files then later downloaded the 4.3.2 sources.  There were several changes in UserModuleBase and UserSettings.asc.vb between the two versions.  It appears that you have version 4.3.2 of UserSettings.ascx.vb which in the cmdUpdate_Click handler calls UserModuleBase.UpdateSettings but have version 4.3.1 of UserModuleBase which does declare an UpdateSettings method.  Apparently this method was not created until 6/27/06 and is declared in version 4.3.2.  For some reason the 4.3.2 version of UserModuleBase.vb either did not install or the compiled library (DotNetNuke.dll) in the Website\Bin folder is an earlier one included with version 4.3.1 rather than the one that would have been included with 4.3.2.

I did have 4.3.1 but I believe that I wiped that folder structure before I went to 4.3.2 and did that as a fresh installation. However, I'm not 100% certain so your explanation could be right.

My solution was to comment out that part of the code because I didn't need it for the debugging that I was doing but I would obviously like to in-comment it when I've updated all the modules in-sync.

Thanks for the help!
