It could help a lot if you can ask your hosting provider...you have the date and time...so they can check their logs.

My problem started yesterday 6PM and I restarted my server to resolve the problem.. I did not restarted only the application (Maybe I was so worried about my server that tried the best solution for a IIS problem...hehe)

Most hosters don't mind to give that information (About server environment)

For me, it looks as a problem with the asp.net itself and not DNN but we must see all possible options and notify the core team if something about DNN is found here.

can you please send me a mail to locopon at tiendaboliviana dot com? I want to exchange some information about this stuff... then we can back to this forum to make the information available to others.

Today, i had same error minutes ago, i am using Ventrian Modules (News Articles & Simple Gallery). And adding some code to HttpHandlers section of web.config helped me.

<!-- This is for Web Resource.axd File -->
      <add verb="GET" path="WebResource.axd" type="System.Web.Handlers.AssemblyResourceLoader" validate="true" />

I had this in my web.config, but the error appeared to be in a "Clear" call that was made by the ventrian modules that actually removed the handler.  I would recommend downloading his newest versions if you have a moment...
