 I am evaluating DNN 4.5.3 for my project.  Can anyone tell me does DNN use enterprise library?

If yes, How do I find which version is being used?Is it safe to upgarde the ENterprise library?

DNN is using the Microsoft Data Application Block for the SQL Data Provider access.

You can check the version based on the dll in the bin directory

As for upgrading it.....I am not sure.

I don't know if your interest in the EntLib is just for data access or some of the other components.  But if it is for data access, I would recommend that you explore the use of the ADO.Net 2.0 features for data access.  This is even easier than using the DAL+ and you can use a resource like this one since developer support for data access in DNN has always been lacking.  And this applies to instances where the data you need to use is in the DNN DB or any other DB.

Granted, this may not be "multi-tiered" by default but to be honest, many projects don't benefit at all from the added complexity.  (You can still make it multi-tiered if you do the extra work, the link above also has examples of that).
