Our server platform runs Visual Studio 2000 - will DNN work with this or must it be VS2005?

You need to take care of your server - this is the first time I hear of Visual Studio 2000, it looks like you got the "Blue Mauritius" of servers

DotNetNuke 3 requires a server running .Net Framework 1.1 And SQL 2000 or MSDE or 2005 or 2008 (even Express editions), to develop modules you need VS 2003

DotNetNuke 4 runs on .Net framework 2.0, same SQL servers and needs VS 2005 or VS 2008 for module development.

Vs2000 does have a web development function however I doubt that the code in DNN3 upwards could be used in Vs2000.DNN4 cannot  be used in VS2000. I assume that the VS2000 on your server is  mainly used for desktop application development. Updating to Vs2003 from Vs2000 is not a hassle and will give you some benefits.
