make sure, default.aspx has been added as default document and the windows account used by ASP.Net (as specified in your IIS Application pool) has full file system permissions for the DNN installation directory and all subfolders.

Thank you for your reply. The default file is Default.aspx and ASP.NET has full permissions. I have set up several DNN sites before without this issue and the strange thing is that if you type Localhost on the local pc the DNN site comes up but it wont work from any remote pc which indicates some local permission outside of the conventional dnn setup. Would really appreciate any assistance to help me find out what it is. I have compared all of the settings with a site I set up a few weeks ago and they all seem to be the same but that one works and this one does not.

if you want to access the site from remote, the domain address needs to be assigned to the IIS website (using host header value) and in DNN you need to add it as additional portal alias.

Thank you so much Sebastian, it was set up correctly in the IIS but the only alias was Localhost. Adding the additional alias did the trick, definitely one to remember for the future.
