While technically the DNN sql provider model was originally designed as a way for more than one sql server to be utilized - its not really the case these days.  The DNN core is very closely tied to MS SQL - and since DNN itself needs to be run on a MS Server and MS SQL Express is free there is not really any great reason for wanting to head down the mysql path.

So the simple answer is no - the more complex answers is - it could be made to if someone wanted to go to the effort of porting the dnn core sql provider scripts to mysql compatibility.  But having said this - there would be little chance of many modules working and no forward support for new versions without redo that process.

To add to what Westa said about MS SQL Express, it's not only free but it is still a very robust database system. With R2 Express you get 10 GB of storage per database (which is plenty for most DNN Installations) and a few limited reporting service features.

Since DotNetNuke is a .net application, MS SQL probably would be your best bet.

I'll see a nice development for MySQL

http://dnnmysql.codeplex.com/

It's still made for DNN 6 but maybe the community will pick it up and fix it for 7 as well.
@JT_Gregory. The stability of MySQL (now a day in MariaDB) and the speed do attempt to keep track for MySQL development. True, Microsoft did a very good job with MSSQL. But as a opensourcer , i'll like it when a good working opensource software can beat the prof.

DNN 7 introduces using PetaPoco micro ORM, I haven't checked out, whether this simplifies creating a different provider or makes it more difficult.
