You have to install DNN4 on the other system, which in your case: copy all the files to the new virtual directory, adjust the folder permissions and web.config, and then do a full sql server back-up and restore. There might be some "back-up" type modules for sale on SnowCovered.com, but I'm not sure how well they work -- especially when you consider that something like this is version specific because it deals with the core database tables.

I'm responding to this, because I'm wondering if the DNN team is working on a better way to deploy DNN sites?

An alternative to that is to create a template of the portal, including module content, from your dev environment, and to destroy your portal on your production server then to re-create the portal on the prod environment. There are a couple downsides to that: if you have some links within your site, you'll have to re-set them because their target will be pointing to the tabIds of your dev environment (there might be some other bugs like this one). Also, if you have many users associated to that portal, you must tel them bye bye; they'll be flushed.

So this method is usefull if you have only one user.

Edit: I must admit that this method is like using a bazooka to kill a spider. I admit that the "support" to migrate a portal or a part of it is lacking. But I do understand that the original purpose of DNN was that: a portal.
