I have over the years always done the classic approach of putting web files and services on a seperate box from the SQL database for all my custom software projects. At least years ago, that was the recommended approach because of the conflicting memory and disk I/O needs.

Anyone ever do performance testing of everything on one box versus two for DNN? Or a whitepaper? What are the pros/cons?

there are no issues running both servers (IIS & SQL) on the same machine, if there are sufficient resources, however to increase performance, separating both is a valid option. SQL Server Express should get at least 1 GB of RAM, a web site may easily require 100-500 MB plus OS.
