I have exceptions like this on my DNN eventlog. I'm using 5.5.0

RawURL: /Nacionales/tabid/6740/NewsId/6713/language/en-US/Default.aspx
AbsoluteURL: /Default.aspx
AbsoluteURLReferrer:
UserAgent: Mozilla/5.0 (compatible; Yahoo! Slurp; http://help.yahoo.com/help/us/ysearch/slurp)
DefaultDataProvider: DotNetNuke.Data.SqlDataProvider, DotNetNuke.SqlDataProvider
ExceptionGUID: 394e023c-e5c3-4f2c-9418-f2be6f39aee5
InnerException: The process cannot access the file 'C:\xxxxxxxxx\DesktopModules\CodingStaff.Comments\License.txt' because it is being used by another process.
FileName:
FileLineNumber: 0
FileColumnNumber: 0
Method: System.IO.__Error.WinIOError
StackTrace:
Message: DotNetNuke.Services.Exceptions.PageLoadException: The process cannot access the file 'C:\xxxxxxxxxxx\DesktopModules\CodingStaff.Comments\License.txt' because it is being used by another process. ---> System.IO.IOException: The process cannot access the file 'C:\HostingSpaces\adn\DesktopModules\CodingStaff.Comments\License.txt' because it is being used by another process.at

And they raised errors like there:

Faulting application w3wp.exe, version 7.0.6002.18005, time stamp 0x49e023cf, faulting module kernel32.dll, version 6.0.6002.18449, time stamp 0x4da47967, exception code 0xe053534f, fault offset 0x0003fc56, process id 0x%9, application start time 0x%10.

This happened after an upgrade to a new server:

Windows Server Entreprise without Hyper-V 32 bits
IIS 7.0.6002.18005
SQL SERVER 10.50.1600.1

That causes the app pool to recycle and it even stop it.... after a day we went back to the old server (changed our DNS) and now I can access the site on this server without any issues or exceptions... even remotely.

I did noticed that the search engine robots where reindexing the new server. In reading and google it I found that exception 0xe053534f
is a "Not handled exception throwed by the code" but it seems to me that DNN is handling it....

 Any ideas?

most often this type of issue (resources competing for file access) is caused by an overly agressive virus scanner or windows indexing services (you might want to exclude the website from the index) -please check your windows event viewer to see if it gives you any useful information
