You can do this in code behind.

ascx:
<asp:Button id="TestButton" onload="TestButton_Load" runat="server" />

ascx.cs:
protected void TestButton_Load(object sender, EventArgs e)
{
Button testButton = (Button)sender;
testButton.Text = Localization.GetString("Term", base.LocalResourceFile);
}

Please note that if you do not add any "extension" to the key, DNN localization searches for "Term.Text". "Term" and "Term.Text" are equal. If you do not have a key called "Term.Text", the thing won't work.
