I created custom roles and assigned them to users from AD and noticed that after certain period of time (~an hour) those roles were removed from users. Any idea?

Do you have role synching selected in the options? If you do then there has to be a corresponding group in the AD.

My custom roles are not a group in the AD. They are roles in DNN.

Do you have role synch turned on in the AD provider settings? If yes, then turn it off and see if that solves the problem.

Yes. and then I turned it off. So far I haven't got any problem.
