OK, let's say I've got everything running like I want it locally (i.e. on my PC). Now, I want to migrate it to a server at a business.

  x  I migrated all the files in the DotNetNuke folder to a folder on the server giving necessary rights to ASPNET user.
  x  I restored the DB to the SQL Server on the server.
  x  I set up the virtual directory in IIS.
  x  I updated the web.config file for the SQL Server connection.

VOILA...works great if I do it locally on the server (i.e. http://localhost/DotNetNuke).

However, how do I set things up for others, from other machines, to connect to the site now? Do I have to create a DNS entry on the server? Do I have to update PortalAlias table and, if so, how? Anything else I'm missing? Is this all documented somewhere?

You probably need to remove the reference to localhost/dotnetnuke in your portal alias's and add the url for the site.  You can do that from the host settings menu or directly in the SQL table.
