That what am I looking forward to. How to code this partial rendering at page load of my module to enable it if the host disabled it from the module definition. Is that possible?

Not sure I follow your question.  The updatepanel can be included in your module like any other control.  (Pretend you were coding this in a separate ASP.NET app).  you are in complete control over how it works.  The only trick is making sure you register the scriptmanager through the DNN framework.
