I believe you have to open the DotNetNuke.sln file under the directory you extracted to from the DotNetNuke_04.08.02_Source.zip you downloaded.  I did not document the steps very well as they wanted it up and running "yesterday" as usual.  After it opens in VS2005 I believe you need to set the default.aspx as the start page for the project.

As soon as I get DNN working in the production environment I want to go back and do some installs from the beginning and document the procedure completely as it needs to be done in our environment.  Of course you know what kind of priority that normally gets assigned.

Racked my brain on this for hours....NETWORK SERVICE had all the permissions but nothing.  But I got this to work:

  1. Remove  (delete) NETWORK SERVICE form the permissions on your DNN virtual directory
  2. On the Virtual Directory Properties "ASP.NET" tab switch it to ASP 1.1 (just go with me here)
  3. Restart the website
  4. Add NETWORK SERVICE on the permissions for your DNN virtual directory
  5. On the Virtual Directory Properties "ASP.NET" tab switch it back to ASP 2.0
  6. Restart the website

This worked for me.  My theory is that when you set the Virtual Directory to "ASP.NET 2.0" is puts a lock on web.config thus preventing a change in permissions...so your change in permissions has to be while the site is set on ASP 1.1.
