On my local computer, using SqlServer 2000, installing 4.0, I solved this using this connection string:

  <!-- Connection String for SQL Server 2000/2005     -->
    <add key="SiteSqlServer" value="Server=51366300K\LOCALSQL;Database=DotNetNuke;uid=sa;pwd=sa;" />

I would also appreciate some help. I'm getting the same error using Visual Web Developer Express 2005, SQL Server Express 2005 and DotNetNuke version 4.

Are those two lines the only changes that you made to the WEB.CONFIG file, or did you edit anything else, or edit any other lines?

Is your SQL Server 2000 using SQL authentication or local?

Besides the connection string, I commented out the SQL Express stuff (in 2 places).  These are the only changes I made to the default web.config (release.config) installed with DNN 4.0. 

My local SQL Server 2000 uses Windows authentication only, no SQL auth allowed. 

The default server string "(local)" must be changed; my combination of "machine-name\LOCALSQL" did work for me.  Uid and pw also must both be given values and not left as "uid=;" or "pw=;" and asterisks didn't work, nor did any valid SQL user with full permissions, etc. except "sa" and "sa".  Believe me, I tried every combination of the above.

I have the same error when i install dotnetnuke on localhost. You should restart IIS when you change the web.config file.
