I feel your pain John.  I had a disastrous update from 5.6.2 to 6.0 on 1 of 4 sites.  So I'm attempting to rebuild the site from scratch and getting no where with the SQL part.  For a time I was getting the same error as you, but for some reason that has disappeared and now I get:

Connection Error(s):
Index #: 0
Source: .Net SqlClient Data Provider
Class: 20
Number: 2
Message:

Unfortunately I don't get any text in the Message: space.  In the event log on my 2008 R2 64bit server I get this message:

Failed to generate a user instance of SQL Server due to failure in retrieving the user's local application data path. Please make sure the user has a local user profile on the computer. The connection will be closed. [CLIENT: <local machine>]

Like you I've tried everything under the sun as far as suggestions go and I can't get past this.  Very frustrating indeed.

Hey at one point I crashed all 4 of my sites while trying to fix this one site, so it could be worse?

Dean, your issue is different from John's and is permissions related -your database connection is using user instances which need to create a database effectively in an admin context and your user does not have permissions - this can be for a host of reasons e.g. see http://social.msdn.microsoft.com/forums/en-US/sqldatabaseengine/thread/fbf32aa8-2125-4919-be81-3baa399e2e0d/ for some common ones. Id recommend you use conventional sql server databases rather than sql server user instances

After a few more hours of research, thank you google, I came across an obscure post that said to leave the database field blank when at the database screen during install.  Well that worked and the site is up and running.  My only concern at this point is where is the data being stored?  How would I ensure that the site is using the sql express database I created?  Would making the change in web.config fix this?

if you've used user instances it will be in the app_data folder -if you want to connect to it you can do so with the usual sql management tools - but be careful you will cause the database to lock whilst you do this so dont forget to disconnect. If you prefer to use sql server without user instances you can attach the database in sql server management studio, but you will need to update both the connection strings in web.config accordingly - http://connectionstrings.com/sql-server-2008

Well I found the database that it created without specifying the database during install.  It put it in Users\%username%\AppData\Local\Microsoft\Microsoft SQL Server Data\SQLEXPRESS under the name Master.mdf.  What would be involved in moving this database to location where I store all my other database files for DNN?

Can this be easily done within SQL Server Management Studio?
