I had websites running on DNN 4.0 on windows 2000 domain controller with IIS.
I built a new machine running windows 2003, set it up as a domain controller, and added the "application server" role.  SQL Server 2005 is installed and running.  I can poke around in the "DotNetNuke" database and everything looks fine.
I have scoured the web and read / tried several documents on how to make this go, but to no avail.  When I browse to any DNN website, I get a 404 error.  However, I have no idea what it can't find.  There is, true to form for microsoft products, precious little debugging information available, and the log files just show a fetch of default.aspx, but nothing else useful.
I can't be that far off because Rainbow Portal websites work just fine.  I have no idea why DNN is being so stubborn, but I'll bet it's simple and probably somewhat arcane.  Any ideas folks?

Do you get any specific error when browsing to your dnn site?

Did you allow asp.net in the Web Service Extensions?

For both security and performance reasons, running IIS on a domain controller should be done as a last resort. MS has a short video on doing so successfully..You can find it here.

Chris,

I agree with that but taken in account that Small Business Server has been sold over 3 million times and that is a domain controller with IIS installed you don't have a choice if you want to run a small Dotnetnuke site on your own server. Because of my site (www.smallbizserver.net) more and more small business owners see the benefit of running Dotnetnuke on their server. You can agree with that or not, you can find it a bad idea or not. It is reality and we should make the best of it.
