Please check the physical file permissions in the root folder where your DNN Platform is located.  Make sure the application pool user have full permissions, and the IIS User (IUser) have read permissions.  Make sure you have inherit permissions to all files and subfolders!  Check .Net settings and be sure that you use .NET 4.x and you have enabled permissions to execute scripts and executalble files.

Then check you have enabled TCP/IP protocol on your SQL Client and Server protocol Settings.

Restart the IIS.

If you are using the file based SQL Server Connection only one (1) Connection is allowed!  If you want to open the webside you have to disconnect the database file from Visual Studio.
