I had same problem & got it going by following this article

http://technet.microsoft.com/en-us/library/bb232021(EXCHG.80).aspx

& adding the ip of the web server to the ip allow list on the anti spam tab next to receive connectors on the hub transport tab in server configuration.

took all of a couple of minutes

hope this helps

The other option is creating an account on the server to use as the DNN sender, normally an AD account, and use that user email address as login and password.  This way you allow relay for authenticated accounts only and don't open up the system to a possible spoof.
