You can apend ?ctl=login to any page and you will be re-directed to your default login page, which if you've already disabled through other means hopefully won't be an issue.

By removing the login.aspx page completely, would this prevent any type of login?  Do you absolutely need the login.aspx page in order to login or can you pass credentials through the query string?

you need a login page as the parameters need to be passed to the dnn memberrole capability to validate the user (doing this via querystring's would be insanely dangerous). I'd suggest your best option is to create new login control that checks for the user's IP address and only loads if it's within your prescribed range (i.e your internal network). You can then set this as the login page within the site settings.

I have no intention of doing logins via the query string, essentially what I am asking is by removing the login.aspx page, would this prevent someone from logging in to the portal completely?

yes, no one will be able to log in include admin and host users.
