No host header is needed, and assuming your website is already configured to serve ASP.NET 3.5, you should have nothing else to do.  Your access URL would be http://{Host}.{Domain}.{TLD}/{Virtual....  The IP address may replace the host.domain.tld portion, but since you're in a virtual directory that has to stay in the path.

Have you had specific problems when you tried to run the web site?

I did not have any specific problems running the site. But, I did not know how to get to it from outside my server (did not know what address to use since it siad "localhost/blah bla/cotnetnuke".

if you install from the server using localhost, you need to add external address manually from within DNN in Host :: Portals menu item: select your portal and add the address in Portal Alias section. You DO need to setup the IIS web site properly to run DNN. This includes host headers and some other settings. If DNN is installed in web site root, you don't need to add a redirect (e.g. to /blahbla/dotnetnuke.
