I am interested in implementing a "point" system for users, where they gain or loose points based on some set of criteria. Ideally this number would be tied to their profile.

I can easily add an integer property "Points" to their profile, but how do I modify this property either through C#, or ASP.NET?

you may either use a dedicated module like on this site (see Community > Community Recognition) or use user profile to store just the total point number, but make sure, the user is not able to edit this property. You may use DotNetNuke.Entities.Users.Profile class to modify the value
