I’m running dnn6.1 on win2008 64bit. I’ve Installed this portal 2 months ago & everything works fine untile a sudden exception occurred today -I can’t logon to my dnn site as host user. All other users are working fine.

Here is the error that I'm getting under google chrom:

http://mydomain/Default.aspx?tabid=56&error=Object+reference+not+set+to+an+instance+of+an+object.&content=0

here is the event log from the database - same set of error entries each time I try to logon as host user

Check to make sure your AppPool in IIS has the same permissions as your web folder and the service running your database.
After that.. check that your SuperUser account is still set to a value of 1 in the IsSuperUser column

SELECT * FROM [Yourdatabasenamehere].[dbo].[Users]
Where Username = 'host'

I'm guessing, without seeing the real exception details, is that you've lost your database connection either because the permissions don't match, the database string is inccorrect in web.config, or the machine/server is not reachable from you DotNetNuke installation
